The fundamental idea behind a cryoEM single particle analysis is that one can purify and then image a large number of essentially identical biological complexes, and then use iterative computational methods to combine the two-dimensional (2d) projection images from the TEM into a three-dimensional (3d) model that is self-consistent (i.e., the vast majority of the original images correspond to particular projection angles through the 3d model) and reliable (i.e., the final model is robust and independent of things like the starting model and image processing approaches).
